چکیده

Abstract Background and Aims Chronic kidney disease (CKD) is associated with high cardiovascular mortality rate, especially because of altered glucose metabolism insulin resistance. Fibroblast growth factor 19 (FGF19), an intestinal postprandial hormone, has been shown to improve metabolic disturbances In CKD, secretion FGF19 seems blunted in hemodialyzed patients but the role regulation this population unknown. The aim our study ascertain if can homeostasis a mouse model CKD induced by 5/6 subtotal nephrectomy. Method Four weeks post-surgery, mice or sham were treated during 18 days subcutaneous injections placebo human recombinant (0.01mg/kg body weight). effect on was estimated vivo tolerance test (GTT, 1g/kg/body Results Fasted hyperglycemia observed groups compared (151.3 +/- 10.6mg/l 170.7 10.9mg/L, p < 0.001 mice, respectively) tended fasting glycemia (158.9 18mg/L, p<0.09). GTT area under curve (AUC) significantly decreased CKD-FGF19 group (P 0.0001) control = 0.0063). Urea blood level higher (9.6 1.4 mmol/L 25.5 5.6 not affected treatment (24.8 5.8 mmol/L, 0.91). Conclusion Our results show that could contribute intolerance suggest hormone be novel therapeutic target for reducing CKD. comprehension molecular cellular events through which exerted its benefits needs however further studies.

Caveolin gene transfer improves glucose metabolism in diabetic mice.

Caveolin, a member of the membrane-anchoring protein family, accumulates various growth receptors in caveolae and inhibits their function. Upregulation of caveolin attenuates cellular proliferation and growth. However, the role of caveolin in regulating insulin signals remains controversial. Here, we demonstrate that caveolin potently enhances insulin receptor (IR) signaling when overexpressed ...

Therapeutic vaccine against DPP4 improves glucose metabolism in mice.

The increasing prevalence of type 2 diabetes mellitus is associated with a significant economic burden. We developed a dipeptidyl peptidase 4 (DPP4)-targeted immune therapy to increase glucagon-like peptide 1 hormone levels and improve insulin sensitivity for the prevention and treatment of type 2 diabetes mellitus. Immunization with the DPP4 vaccine in C57BL/6J mice successfully increased DPP4...

1-Deoxynojirimycin Alleviates Liver Injury and Improves Hepatic Glucose Metabolism in db/db Mice.

The present study investigated the effect of 1-Deoxynojirimycin (DNJ) on liver injury and hepatic glucose metabolism in db/db mice. Mice were divided into five groups: normal control, db/db control, DNJ-20 (DNJ 20 mg·kg(-1)·day(-1)), DNJ-40 (DNJ 40 mg·kg(-1)·day(-1)) and DNJ-80 (DNJ 80 mg·kg(-1)·day(-1)). All doses were treated intravenously by tail vein for four weeks. DNJ was observed to sign...

Acute Fornix Deep Brain Stimulation Improves Hippocampal Glucose Metabolism in Aged Mice

Background A beneficial memory effect of acute fornix deep brain stimulation (DBS) has been reported in clinical studies. The aim of this study was to investigate the acute changes in glucose metabolism induced by fornix DBS. Methods First, the Morris water maze test and novel object recognition memory test were used to confirm declined memory in aged mice (C57BL/6, 20-22 months old). Then, f...
